The Host Defense Group of the Laboratory of Respiratory Biology, Division of Intramural Research, National Institute of Environmental Health Sciences, headed by Michael B. Fessler, MD now enters its 18th year at the NIEHS. Publications since the last annual report include:

1. A collaborative report on the mechanism by which cockroach allergen Bla g 1 elicits a pro-inflammatory response in epithelial cells.
2. A collaborative report of a novel CD301b+ subset of dendritic cells in the mouse lung that is programmed by the cytokine granulocyte-macrophage colony-stimulating factor
3. A senior-author pre-print on the role of the membrane protein Flotillin-2 in regulation of T cell receptor signaling strength and T cell functions
4. A collaborative report showing that ozone inhalation in mice does not recruit monocyte-derived alveolar macrophages into the lung and that tissue-resident macrophages instead mediate clearance of apoptotic cells in the lung.
5. A senior-author report demonstrating that sterol synthesis is required to support the innate immune response in cells, and that failure of this underlies the immunodeficiency of the rare cholesterol synthesis disorder, Smith-Lemli-Opitz syndrome.
6. A collaborative review article on the role of oxysterols in host defense
7. A collaborative paper showing that oxidized low-density lipoprotein reprograms cellular metabolism in macrophages
8. A senior-author report showing that the protein IRGM1 supports host defense against intracellular bacteria by suppressing type I interferon induction.
9. A collaborative report that DNASE1L3 enhances antitumor immunity in mice.
10. A senior-author publication reporting that 25-hydroxycholesterols may worsen SARS-CoV-2-mediated pathogenesis in mice during pneumonia.

Additional accomplishments since our last report include invited presentations by Dr. Fessler at Cedars-Sinai Medical Center, University of Alabama Birmingham, Massachusetts General Hospital, Beth Israel Deaconess Medical Center, and at the N.C. Visiting Pulmonary Scholars Symposium. Dr. Fessler was also promoted to Deputy Editor for the American Journal of Respiratory Cell and Molecular Biology.
