The overarching goal of this project is to study the immune response to pathogens at the monoclonal level to increase our understanding of basic biology and develop new tools to combat disease. We are currently focusing our efforts on human monoclonal antibodies against two pathogens, Plasmodium falciparum and SARS-CoV-2. These projects have started recently, so most of our findings are still preliminary at this stage. For SARS-CoV-2, we have screened plasma from >100 acutely infected and convalescent COVID-19 individuals for neutralization of authentic SARS-CoV-2. The top neutralizers were selected for antibody isolation and we have isolated a panel of monoclonals against the SARS-CoV-2 spike protein. Characterization of these antibodies is currently underway, and our hope is to identify antibodies that can potently neutralize the virus and delineate specific epitopes that may be suitable vaccine candidates. For P. falciparum, we are in the process of setting up high-throughput assays to screen for antibodies against various stages of the life cycle, including sporozoites, blood stages and sexual stages.
