Using CryoEM can provide great insight into a protein's structure and function. Proteins under 50 kDa in size are difficult to image by standard CryoEM procedures. The legobody system helps to circumvent this problem through the generation of a large, stable protein complex composed of four proteins. Using a single-domain antibody as an adaptor allows the target protein of interest to bind to the Legobody complex. We believe that incorporating the NCATS' sdAbs into the legobody system will help accelerate future CryoEM studies and drug development efforts here at NCATS.
