Principal Investigator: William Elkins Organization: NATIONAL INSTITUTE OF ALLERGY AND INFECTIOUS DISEASES Fiscal Year: 2022 Award: $28,533 Funding agency: National Institute of Allergy and Infectious Diseases COVID 19 related projects: Humanized animals: Using CRISPR/cas9 mediated gene KI via specific gRNAs and ODNTs, several lines have been created with the human ACE2 mutations individually in exons #2, exon #3 and exon #9. In the second stage, the sperm from these lines was used to create embryos that were targeted for second exon and created lines with dual mutations in exon#2 and exon#9 which will be used to create triple mutants humanized animals. COVID-funding: MGGM was awarded additional $36,000 for the creation of humanized ACE2 NOD/ShiLtJ mice as model for underlying conditions in humans. Revised proposal was submitted for humanizing ACE2 gene of the Thicket rat as a test model in collaboration with Dr. Patrick Duffy and Dr. Heather Hickman. Novel ES cell lines created from the Thicket rat will be utilized for creating humanized animals via tetraploid mouse embryo complementation as placenta donor Terms: <2019 novel corona virus><2019 novel coronavirus><2019-nCoV><ACE2><Animal Model><Animal Models and Related Studies><Animals><Award><COVID><COVID-19><COVID-19 virus><COVID19><COVID19 virus><CRISPR><CRISPR approach><CRISPR based approach><CRISPR method><CRISPR methodology><CRISPR technique><CRISPR technology><CRISPR tools><CRISPR-CAS-9><CRISPR-based method><CRISPR-based technique><CRISPR-based technology><CRISPR-based tool><CRISPR/CAS approach><CRISPR/Cas method><CRISPR/Cas system><CRISPR/Cas technology><CRISPR/Cas9><CRISPR/Cas9 technology><CV-19><CV19><Cas nuclease technology><Cells Placenta-Tissue><Clustered Regularly Interspaced Short Palindromic Repeats><Clustered Regularly Interspaced Short Palindromic Repeats approach><Clustered Regularly Interspaced Short Palindromic Repeats method><Clustered Regularly Interspaced Short Palindromic Repeats methodology><Clustered Regularly Interspaced Short Palindromic Repeats technique><Clustered Regularly Interspaced Short Palindromic Repeats technology><CoV disease><CoV-2><CoV2><Collaborations><Common Rat Strains><Complement><Complement Proteins><ES Cell Line><Embryo><Embryonic><Embryonic Stem Cell Line><Exons><Funding><Gene-Modified><Genes><Genetic Alteration><Genetic Change><Genetic Engineering><Genetic Engineering Biotechnology><Genetic Engineering Molecular Biology><Genetic defect><Guide RNA><Human><Individual><Investigators><Mediating><Mice><Mice Mammals><Modeling><Modern Man><Murine><Mus><Mutation><NIAID><National Institute of Allergy and Infectious Disease><Normal Placentoma><Placenta><Placenta Embryonic Tissue><Placentome><Rat><Rats Mammals><Rattus><Recombinant DNA Technology><Research Personnel><Researchers><SARS corona virus 2><SARS-CO-V2><SARS-COVID-2><SARS-CoV-2><SARS-CoV2><SARS-associated corona virus 2><SARS-associated coronavirus 2><SARS-coronavirus-2><SARS-related corona virus 2><SARS-related coronavirus 2><SARSCoV2><Severe Acute Respiratory Coronavirus 2><Severe Acute Respiratory Distress Syndrome CoV 2><Severe Acute Respiratory Distress Syndrome Corona Virus 2><Severe Acute Respiratory Distress Syndrome Coronavirus 2><Severe Acute Respiratory Syndrome CoV 2><Severe Acute Respiratory Syndrome-associated coronavirus 2><Severe Acute Respiratory Syndrome-related coronavirus 2><Severe acute respiratory syndrome associated corona virus 2><Severe acute respiratory syndrome corona virus 2><Severe acute respiratory syndrome coronavirus 2><Severe acute respiratory syndrome related corona virus 2><Sperm><Spermatozoa><Testing><Wuhan coronavirus><angiotensin converting enzyme 2><angiotensin converting enzyme II><corona virus disease><corona virus disease 2019><coronavirus disease><coronavirus disease 2019><coronavirus disease 2019 virus><coronavirus disease-19><coronavirus disease-19 virus><coronavirus infectious disease-19><gRNA><gene modification><genetically engineered><genome mutation><hCoV19><model of animal><model organism><mouse genetics><mutant><nCoV2><novel><sperm cell><therapeutic target><viral transmission><virus transmission><zoosperm>
