Monoclonal antibody isolation and characterization for COVID-19 Effort

Principal Investigator: Rosemarie Mason Organization: NATIONAL INSTITUTE OF ALLERGY AND INFECTIOUS DISEASES Fiscal Year: 2024 Award: $17,731 Funding agency: National Institute of Allergy and Infectious Diseases We have isolated multiple SARS-CoV-2-specific monoclonal antibodies (mAbs) from SARS-CoV-2 convalescent donors from study VRC200. More than 25 mAbs isolated and characterized were neutralizing against SARS-CoV-2 with several mAbs showing cross-neutralization of multiple variants of concern (VOC) as well as SARS1, and other SARS1- and SARS2-related coronaviruses. We are drafting a manuscript to describe the isolation, characterization and high-resolution structures of 5 pan-coronavirus neutralizing mAbs. Structural analysis revealed that 2 of the mAbs disrupt/dissociate the spike trimer and target a relatively conserved epitope.
